Sinopse: Convencido que o seu problemático irmão, Lincoln Burrows (Dominic Purcell), foi mal condenado pelo sensacionalista assassinato do irmão da vice-presidente, que o levou para o corredor da morte da Prisão Estatal de Fox River, o desesperado Michael Scofield (Wentworth Miller) tentou um ousado roubo a um banco para ficar preso com Lincoln. Michael, um engenheiro civil com acesso às plantas da prisão, planeou e organizou um plano super elaborado para libertar Lincoln ao tentar a mais arriscada fuga da prisão (Prison Break). Michael junta-se ao seu companheiro de cela Fernado Sucre (Amaury Nolasco), ao ex-chefe da Máfia Jonh Abruzzi (Peter Stormare), ao pedófilo assassino “T-Bag” (Robert Knepper), à pessoa com os conhecimentos certos “C-Note” (Rockmond Dunbar) e a Charles Westmoreland (Muse Watson), um homem que Michael acredita ser o famoso D.B. Cooper, para prosseguir com o seu plano. No lado de fora, a unica aliada de Michael é a sua advogada e ex-namorada de Lincoln, Veronica Donovan (Robin Tunney). Entre as personagens secundárias encontram-se a Dra. Sarah Tancredi (Sarah Wayne Callies), o director Pope Henry (Stacy Keach) e o Guarda Prisional Bellick (Wade Williams).
